During 2022\/23 season, samples collected from influenza-like-illness patients as of July 24, 2023 were as below:<\/p>\n

260 samples were collected at the first visit, 86 samples at the second visit, 54 samples after baloxavir treatment from Japan (Table 1).<\/p>\n

The results of the detection of PA\/I38T substation by cycling probe real-time PCR are shown below (Table 2).<\/p>\n

The percentage of patients with PA\/I38T substitution was:
\nIn A(H1N1)pdm09, no PA\/I38T variant in pre- and post-treatment samples was detected out of 6 samples (0.0%). In A(H3N2), among pre-treatment samples 2 PA\/I38T variant (0.8%) was detected out of 231 samples. Among post baloxavir-treatment clinical samples, 3 PA\/I38T variants (5.6%) were detected out of 53 samples. If the frequency is calculated based on RT-PCR-positives at the second visit, it became 14.3% (3 PA\/I38T variants out of 21 RT-PCR-positive samples at the second visit).<\/p>\n

In unsubtyped influenza A, pre-treatment samples were 0 positive (0%) PA\/I38T variant out of 9 specimens.<\/p>\n

Influenza B was not detected.<\/p>\n

[PA gene analysis]<\/p>\n

In A(H3N2), three I38T substitutions (7.1%) out of 42 paired baloxavir-treated clinical sample was detected by next-generation sequencing (NGS) (Table 3). If the denominator is based on 9 samples with successful NGS in the baloxavir-treated group, the frequency of PA variant was 33.3%.<\/p>\n
